The Itar-Tass quoted police source as saying that the governor was assassinated by a pistol with muffler when he was walking out of his regional office in Moscow on Novy Arbat Street, one of the capital's busiest thoroughfares.
Two assailants fled in a car after discarding their weapons, the agency cited eyewitness as saying.
President Vladimir Putin has instructed thorough probe into the accident, said the presidential press service.
Tsvetkov, 54, has been working as the governor of the gold-rich Magadan region since 1996.
